| 摘 要： | 为鉴定和验证ROCK1和CD40的相互作用,探索ROCK1在CD40通路中可能发挥的作用,应用HD质谱分析ROCK1和CD40的相互作用,用免疫共沉淀实验验证其相互作用, 用ROCK1抑制剂研究ROCK1对CD40通路的影响. 结果ROCK1与CD40存在相互作用;并参与CD40介导的信号通路.说明ROCK1能与CD40发生相互作用,为CD40信号通路的研究提供了新的线索.

| Abstract: | Objective: To identify and validate the interaction between ROCK1 and CD40 in order to explore the function of ROCK1 in CD40 pathway. Methods: The HD MS was applied to screen the protein interacted with CD40 and ROCK1 as an important protein was focused on. The interaction was demonstrated by co-immunoprecipitation assays. The influence of ROCK1 in CD40 pathway was researched by ROCK-specific inhibitor. Results: ROCK1 was conformed to interact with CD40 and involved in CD40 pathway. Conclusion: ROCK1 interacts with CD40, which may provide an insight on the regulation of CD40 pathway |
