摘    要:植物类蛋白激酶ABC1家族(Activity of bc1 complex)在植物生长发育及响应非生物胁迫中扮演着重要的作用。我们克隆并研究了ABC1家族中一个的基因AtOSAK1。半定量RT-PCR及启动子驱动GUS报告基因分析显示,AtOSAK1基因的表达无组织特异性,并且主要受甲基紫精(MV)的诱导表达。AtOSAK1-GFP亚细胞定位显示,AtOSAK1是一个定位在叶绿体中的蛋白。不同浓度MV处理条件下,过表达AtOSAK1植株的种子萌发率和存活率比野生型(Col-0)和突变体osak1高。这些研究表明,AtOSAK1基因参与了抗氧化胁迫的响应.

Abstract:A typical kinase family, ABC1 (the activity of bc1 complex) genes play essential roles in plant growth, development and responses to be environmental stresses. We cloned and characterized a gene of the ABC1 family in Arabidopsis. Semi-quantitative RT-PCR and GUS staining analysis revealed that it was constitutively expressed and mainly induced under methyl viologen (MV) treatment. AtOSAK1-GFP fusion analysis showed that AtOSAK1 is localized in chloroplasts. Transgenic plants overexpressing AtOSAK1 showed higher rates of seed germination and seedling survival to MV stress than that of the Col-0 and the knock-out mutant osak1. These results suggested that AtOSAK1 may be involved in response to oxidative stress in Arabidopsis.
