摘    要:以油菜的子叶柄作为转化的受体材料,经农杆菌感染和共培养,在含卡那霉素的筛选培养上诱导产了大量的不定芽,经生根培养,获得了再生植株,通过PCR扩增和基因组Southern杂交分析,证明其中5株为转基因植株,对转基因植株进行抗虫性检测,有2株表现出较好的抗虫性。

Abstract:WT5"BZThe cotyledon petiols of oilseed rape WTBX(Brassica napus)WTBZ were used as recipient of transformation. After being infected and co-cultured with WTBXAgrobacterium tumefaciens,WTBZcotyledon petiols produced a lot of adventitious buds on selection medium supplemeted with kanamycin. Kanamycin-resistant plants were then obtained from the adventitious buds by rooting culture. DNA Southern blot hybridization and polymerase chain reaction showed that 5 of the plants were transgenic when probed with the insect-resistant gene. These transgenic plants exhibited different insect-resistant response when they were subjected to leaf feeding experiments, but two of them displayed a good resistance to pest insect larvae.
