绿色荧光蛋白基因在大黄鱼体内的表达

摘    要:以绿色荧光蛋白基因为报告基因,以pIRESnco为载体质粒,制备含有报告基因的质粒,以肌肉注射的方式导入大黄鱼体内,每隔一周用荧光显微镜观察绿色荧光蛋白基因在鱼体不同部位的表达情况。结果表明,绿色荧光蛋白基因可以大黄鱼体内得到表达,表达时间高达28d以上。绿色荧光蛋白基因不仅可以在肌肉注射部都组织细胞得到表达,而且在其它部位的肌肉、肝脏、肾脏和心脏等组织中也有表达,但表达水平低于肌肉注射部位组织。

Abstract:Plasmid DNA pIRESneo containing green fluorescent protein gene was injected into the skeletal muscle of Pseudosciaena crocea, and the expression of green fluorescent protein gene in different tissue of these tested fish was weekly examined by fluorescent microscope after injection. The results showed that the expression of green fluorescent protein gene in P. crocea lasted as long as 28 days postinjection. Expression of injected DNA was found not only in the muscle cells at the injection site, but also in the muscle cells far from the injection site and the other organs such as liver, heart and kidney. The expression level was higher in the muscle cells at the injection site than those in other tissues.
