长豇豆品种资源的RAPD分析

摘    要:从30个随机引物中筛选出23个有效引物,共得到140条多态性带.实验平均每个引物扩增6.13条带,多态性条带数为6.09条.平均每个品种显示多态性扩增带64.35条(变幅为26~82),其变异系数为15.62%;平均每条带出现的品种次数为18.39个(变幅为1~39),频率为0.46(变幅为0.30~0.98),其变异系数为71.45%.品种间RAPD扩增带数目有别,其位点变异更大,说明供试品种间DNA序列存在不同程度差异,为根据DNA指纹有效判别品种间遗传差异提供了可能性.根据长豇豆品种DNA分子RAPD系统树图,可将长豇豆品种分为5个品种群,可弥补根据形态性状异同来判别品种的不足,揭示了由于自然和人工选择可遗传性状变异积累、地理隔离歧化和人工杂交强化基因重组等共同作用而形成的品种基因组差异的现实,也启示着品种亲缘关系确定和品种选择选配在成功改良品种中的重要性.

Abstract:Screensandobtains23 valid primersfrom30 radomprimers,obtains140 polymorphic(amplitude of variation is 1~39),frequency is 0.46(amplitude of variation is 0.30~0.98),the coefficient of variation is 71.45%.The RAPD amplified bands number has difference among sorts,the variation of site is more different,this suggests that DNA sequence of the experiment sorts have difference on different degree,provide the possibility to identify the genetic variation of different sorts according to DNA fingerprint.According to tree map of RAPD of yardlong been cultivar,the sorts of the bean can be divided to 5 groups,this can compensate the defficiency of identifing the sorts by shape.The results discover that because of the accumulational variation by nature or artificial elimination,geographical isolation and artificial genetic recombination,the genome of different sorts has variation,and suggest the importance to reform the sort quality by the blood relationship ofbands.In the experiment,averagely every primer are amplified 6.13 bands,the polymorphic bandsare 6.09.Averagely every sort show polymorphic amplified bands 64.35(amplitude of variation is 26~82),the coefficient of variation is 15.62%;the sort time of every band averagely is 18.39 different sorts.
