摘    要:以荷花花瓣为材料,在比较CTAB-LiCl法和Trizol法的基础上,针对荷花花瓣富含多糖、多酚的特点,在RNA提取过程中加入去多糖步骤,改良了CTAB-LiCl法.结果表明,改良CTAB-LiCl法能有效地去除多糖,提取到的RNA 28 SrRNA亮度约为18SrRNA的二倍,A260/A280比值为1.98,A260/A230比值为2.36,RNA产率为685.3ng/μL,说明利用改良CTAB-LiCl法从荷花花瓣中提取的RNA质量好、产率高、完整性好.

Abstract:Two methods,CTAB-LiCl and Trizol,were used to extract total RNA from petals of lotus,and CTAB-LiCl was accordingly modified in extracting the total RNA in petals of lotus by integrating a polysaccharide-removing step in correspondence with the fact that lotus was rich in polysaccharides and polyphenol.The results showed that the modified CTAB-LiCl was capable of efficiently removing polysaccharides,and that of the RNAs extracted by the modified CTAB-LiCl,the lightness of 28 s rRNA was two times as much as that of 18 S Rrna, the value of A260/A280,A260/A230 was1.98,2.36,and the RNA yield reached 685.3 ng/μL,which explained that the modified CTAB-LiCl could extract high-quality and high-complete RNA at a higher productivity.
