碱性蛋白酶交联酶晶体的制备及稳定性

摘    要:研究了用透析平衡法制备碱性蛋白酶晶体的过程,在硫酸铵饱和度为25%的条件下获得了符合要求的碱性蛋白酶晶体,然后采用戊二醛进行交联,制备出了交联酶晶体,并对其在高酸碱度、高温、有机溶剂或有机溶液中的稳定性进行了研究.实验结果表明,交联酶晶体有较高的稳定性.

Abstract:This paper studies the preparation of crystals of alkaline protease by dialysis equilibrium. Crystals of alkaline protease are grown from 25% saturated ammonium sulfate and cross_linked with glutaraldehyde to produce cross_linked enzyme crystals. Also dealt with is its stability in different environments, including high pH extremes and temperature, with the exposure to organic solvents or aqueous_organic mixtures. The result demonstrates that the stability of alkaline enzyme in form of CLECs is high.
