罗非鱼无乳链球菌LAMP快速检测方法的建立

摘    要:针对无乳链球菌高度保守的表面免疫相关蛋白基因,采用环介导等温扩增技术(LAMP),设计4条特异性引物,优化反应体系,建立罗非鱼无乳链球菌快速诊断方法.研究结果表明,LAMP方法检测灵敏度达到30 CFU·m L-1,且对2株罗非鱼源无乳链球菌均能扩增出特异性片段,而对14株非无乳链球菌均未扩增出相应片段.建立的LAMP检测方法具有高度的特异性和灵敏性,反应在1 h内完成,为罗非鱼无乳链球菌病的快速诊断提供了一种重要的技术手段.

Abstract:To develop a technique for detecting the surface immunogeenic protein(sip) gene of Streptococcus agalactiae using a DNA amplification method designated loop-mediated isothermal amplification (LAMP) . The rapid detection method was established by using optimum reaction system with four specially primers of the target. Results showed that the method had perfect specificity,as 2 strains of Streptococcus agalactiae were able to amplify specific fragments,while 14 strains of non- Streptococcus agalactiae were not.The amplification can be obtained in 1 hour with the sensitivity low to 30 cfu/ml.The established method povides a better choice for rapid detection of Streptococcus agalactiae.
