小鼠转基因及传代研究

摘    要:综述了湖北省农科院生物技术研究所近年来有关转基因小鼠的主要研究进展。应用原核注射技术,将POMT—PGH、hDAF、pBHSA、pSHSA、PT-HBV1.3、Bcl—xL、hCD59、hCD59 hMCP等8种外源基因,注入并移植4874枚小鼠的受精卵,得到556只小鼠;经PCR和Southem杂交检测,确认原代转基因小鼠108只。基因整合率平均19.4%,转基因效率平均2.2%。应用混合注射的方法得到了转双基因小鼠,双基因共整合率22.2%。表达外源蛋白的转基因小鼠在50%-100%之间。研究了小鼠的超数排卵和影响转基因效率的几个因素。通过转Bcl-xL小鼠与非转基因小鼠连续五个世代的传代交配和检测,研究了转基因小鼠外源基因的遗传规律,表明四只原代小鼠中,只有一只能稳定地将外源基因传递给后代。并非所有的转基因小鼠都具有遗传的稳定性,欲建立小鼠的转基因品系,尚需对原代转基因小鼠进行筛选。

Abstract:The main progresses on transgenic mouse in last several years in Biotech Institute of Hubei Academy of Agriculture Sciences have been summarized. 4874 eggs were microinjected with 8 genes, such as POMT-PGH, hDAF, pBHSA, pSHSA, PT-HBV1.3, Bcl-xL, hCD59. 556 baby mouse are obtained. The average integration rate was 19.4%. The average transgenic efficiency was 2.2%. Bi-gene transgenic mouse a re o btained by mixture gene injection. The integration rate was 22.2%. The transgeni c mouse of expressing foreign protein was 50% to 100%. Author studied super-ovu lation and several factors of effecting transgenic efficiency. They studied the heredity rule of foreign gene on transgenic mouse via continuous mating and diag nosing of five generations on Bcl-xL transgenic mouse with non-transgenic mous e. The result showed that one of four original transgenic mice could stably inhe rit foreign gene to their offspring. Some transgenic mice don't have the heredit y stability. It is necessary to select original transgenic mice for establishing a line of transgenic mice.
