摘    要:研究了pH、温度以及抑制剂与激活剂对从新鲜刺嫩芽中提取出的多酚氧化酶(PPO)活性的影响,及温度对其热稳定性的影响.结果表明:以邻苯二酚为底物,该酶的最适pH为4.4,在pH 4.0-4.8内有较高的稳定性;最适温度为12.5℃,在30℃以上失活;70℃热处理90 s可以钝化PPO的活性;抗坏血酸(Vc),L-Cys对PPO有抑制作用,SDS对PPO有激活作用.

Abstract:The influences of pH, temperature, thermal stability, inhibitors and activators on the enzyme activity of polyphenoloxidase extracted from fresh Aralia elata are studied. With catechol as substrate, the optimum pH of the enzyme is 4.4, and the enzyme activity is higher with pH 4. 4--4.8. The optimum temperature is 12.5 ℃, and the temperature more than 30℃ results in the enzyme deactivation. Heat treating for 90 s at 70 ℃ passivates the enzyme activity. The enzyme activity could be restrained by ascorbate and L-Cys, but be activated by SDS.
