前列腺素E1,锌离子对大鼠感觉神经元的保护作用

摘    要:目的探讨环化酶激活剂(AdenyIate cyclase activator)对周围神经损伤后感觉神经元存活和变性的影响.方法大鼠坐骨神经夹毁模型、分组,术后每d分别局部注射一定剂量的PGE1,hβNGF,共14 d.另一组从术前10 d开始喂以高锌饲料各组术后21 d取背根节(DRG)制片染色,用图像分析法观测PGE1和锌对DRG细胞数、核偏心率和核等圆径的影响,以人胎盘神经生长因子(hβNGF)作阳性对照.结果锌显著降低感觉神经元的死亡率,显著减轻核偏移、抑制核等圆径增大,与NGF组相比,P>0 05.PGE1也明显减少神经元死亡,与夹毁组相比,P<0 05结论神经损伤后锌促进感觉神经元存活和减轻细胞变性的保护作用与NGF无差别.PGE1对感觉神经元的保护作用不及NGF和锌.

Abstract:Objective To explore the effects of adenylate cyclase activator, prostaglandin E1 and Zn 2 ions on the survival and degenerative changes in sensory neurons after peripheral nerve injury.Methods Crush models of rat's sciatic nerve were employed with nerve growth factor as the positive control. Model animals were divided into 5 groups. Immediately following operation different amounts of adenylate cyclase activator, nerve growth factor of sterile saline were locally injected once a day for 14 days. In another group high Zn 2 was fed from the 10 days before operation. On 21 days after operation, dorsal root ganglia were dissected for counting the cell number, nucleus eccentricity and radius of equal cycle nucleus with computer image analysis.Results In the animals treated with PGE1, the numbers of neuron death in dorsal root ganglia reduced significantly, but the effects of reducing nucleus eccentricity and restoring the radius of equal cycle nucleus were not significant. Animals fed with high Zn 2 the increase of neuron survival numbers in dorsal root ganglia were even more significant, and less than that treated with nerve growth factors ( P >0.05), and the changes in nucleus eccentricity and the radius of equal cycle nucleus were also markedly lessened( P <0.05).Conclusion There was a prominent protective effect of activator on neurons in DRG. The effects of PGE1 were less marked than that of nerve growth factor. But the effect of Zn 2 was similar to that of it.
