大蒜拮抗性内生细菌的筛选

摘    要:本实验以市面销售大蒜作为实验材料,通过表面消毒,利用粉碎机粉碎方法得到植物组织原液的方法,由平板涂布法分离得到药用植物组织内的内生细菌或者直接在无菌环境中取大蒜组织块在PDA培养基或NA培养基直接进行培养,通过不断地分离纯化提取得到的较纯植物内生细菌作为目标菌,以大肠杆菌(Escherichia coli),耐甲氧西林金色葡萄球菌(methicillin resistant Staphylococcus aurous,MRSA),金黄色葡萄球菌(Staphylococcus aurous)、枯草芽孢杆菌(Bacillus subtitles)作为指示菌进行内生菌抑菌试验,由内生细菌对不同病原菌的抑菌圈大小的比较筛选出抑菌最强的内生细菌。研究结果表明:大蒜中有大量的内生细菌,分离得到16株内生细菌,16株菌对4种以上病原菌均有不同程度的抑制作用。其中DS1、DS2和BS2对四种病原菌的拮抗能力均较强,BS2对四种靶标菌的相对抑菌率在200%以上。

Abstract:we use market sales garlic as experimental material of this experiment,through the surface disinfection mill grinding method to get plant tissue concentrate.Through the plate coating method to separate entophytic bacteria from garlic tissue on the PAD or NA medium.Antagonistic strains against to Staphylococcus anreus,Escherichia coli,Bacillus subtilis,methicillin-resistant Staphylococcus aureus (MBSA)were screened.The experimental results showed that there were a lot of endophyfic bacteria isolated from garlic. Sixteen strains isolated from garlic have varying degrees of inhibitory effect on the four target bacteria,inhibitory effect of three strains (DS1,DS2 and BS2)was very slgnificant,the relative inhibitory rate of BS2 over 200%.
