跨膜蛋白超家族TM9 SFI和炎性反应介质在颅内动脉瘤壁形成和破裂中的作用分析

摘    要:目的探讨跨膜蛋白超家族的TM9SFl和炎性反应介质AZUl在动脉瘤形成和破裂中的作用.方法选取行颅内动脉瘤夹闭术的患者40例,术中取患者头皮撕裂的颞浅动脉为对照组,手术完成后取上端全层血管壁为研究组.应用免疫组化和Wsetern blot法分析TM9SFl和AZUl在破裂动脉瘤组织和颞浅动脉组织的表达和含量.结果免疫组化染色显示:对照组TM9SFl和AZUl呈阴性或弱阳性表达,研究组呈阳性或强阳性表达;研究组TM9SFl,AZUl染色分级为1.90±0.20,2.90±0.30,明显高于对照组的0.70±0.20,0.80±0.10,差异具有统计学意义(P0.05).Wsetern blot显示:研究组TM9SFl表达量为0.95±0.43,明显高于对照组的0.41±0.15,差异具有统计学意义(P0.05);研究组AZUl表达量为1.16±0.48,明显高于对照组的0.39±0.13,差异具有统计学意义(P0.05).结论 TM9SFl和AZUl高表达参与了颅内动脉瘤形成和破裂过程中的血管壁蛋白降解和炎性反应机制.

Abstract:Objective To investigate the role of transmembrane protein superfamily protein TM9 SFl and inflammatory reaction medium AZUl in the formation and rupture of intracranial aneurysm. Method 40 patients accepted intracranial aneurysm clip occlusion in our hospital were enrolled in this study. Superficial temporal artery of the patients during the operation was taken as the control group,and the upper total vascular wall was taken as the research group. The expression and content of TM9SFl and AZUl in ruptured aneurysm and superficial temporal artery were analyzed by Immuno histochemistry and Wsetern blot method. Results The results of Immuno histochemical staining showed that TM9 SFl and AZUl were negative or weakly positive expression in the control group,and showed positive or strong positive expression in the study group. The dying grades of AZUl and TM9SFl in the research group was 1. 90 ± 0. 20,2. 90 ±0. 30,respectively,significantly higher than in the control group with 0. 70 ± 0. 20,0. 80 ±0. 10,respectively(P<0. 05). Wsetern blot showed that the expression level of TM9SFl with 0. 95±0. 43 in the study group was significantly higher than that of the control group with 0 . 41 ± 0 . 15 ( P<0 . 05 ) . The expression of AZUl with 1 . 16+0 . 48 in the study group was significantly higher than that of the control group with 0. 39±0. 13(P<0. 05). Conclusion The high expression of TM9 SFl and AZUl is involved in the mechanism of protein degradation and inflammatory reaction in the process of the formation and rupture of intracranial aneurysms.
