SD大鼠肝癌演进中端粒酶活性的动态变化

摘    要:探讨端粒酶活性在肝癌演进中的变化,用3'-甲基-4-二甲基氨基偶氮苯(3'-Me-DAB)作诱变剂,建立SD大鼠肝癌变动物模型.自灌胃之日起,分别在2、4、6、8、10、12、14、17w分批处死大鼠,取其肝脏,进行病理分级,然后用TRAP-ELISA法检测肝癌变各时期端粒酶活性,结果显示:在轻度炎性反应期(LI)端粒酶活性高于对照组,呈极显著差异(P<0.01);重度炎性反应期(HI)端粒酶活性有所下降,但仍高于对照组,且呈显著性差异(P<0.05);至腺瘤样增生(NH)和肝内胆管细胞癌(CCC)时端粒酶活性再次升高,与对照组相比呈极显著差异(P<0.01).提示端粒酶活性增高可能是肝癌变进程中的重要事件,与肝癌的发生发展密切相关.

Abstract:To discuss the dynamic changes of telomerase during the hepatocarcinogenesis in SD rats,animal models of every pathologic stage are established by 3’-methyl-4-dimethylaminoazobenzene(3’-Me-DAB) gavage.Animals are sacrificed respectively at 2,4,6,8,10,12,14 and 17 weeks from the first day of gavage and the livers are harvested.After pathological grading,TRAP-ELISA has been used to explore the changes of telomerase in the whole process of carcinogenesis.Results show the following: In light inflammation(LI),the expression of telomerase is higher and the difference is significant compared with the control group(P<0.01);To heavy inflammation(HI),the expression of telomerase is gradually decreased but is still higher than the control group(P<0.05);To nodus hyperplasia(NH) and cholangiocellular carcinoma(CCC),the expression of telomerase gradually increase again and there are significant differences between the induced and the control group(P<0.01).The result suggests that the increased activity of telomerase may be very important in hepatocarcinogenesis,so telomerase is closely related to the development of liver cancer.
