| 摘 要： | 采用RT-PCR方法克隆辣椒八氢番茄红素脱氢酶(PDS)基因部分序列,并构建病毒诱导基因沉默(VIGS)载体pYL156-PDS,经农杆菌介导侵染辣椒叶片,采用表型观察、半定量RT-PCR方法评价构建的VIGS体系的沉默效果.结果表明,辣椒新生叶片呈现明显的光漂白现象,且叶片内源PDS基因表达量明显低于对照.研究证明,辣椒VIGS体系已被成功构建,为规模化验证辣椒基因功能提供了技术平台.

| Abstract: | The Capsicum frutescens var phytoene desaturase(PDS) gene fragment was obtained by using RT-PCR and the virus-induced gene silencing(VIGS) vector pYL156-PDS was constructed.The silencing effect of VIGS system was evaluated by using phenotype observation and semi-quantitative RT-PCR method after Capsicum frutescens var leaves was infected by Agrobacterium GV3101.The newly born leaves showed obvious photobleaching symptom,and the leaves endogenesis PDS gene expression were significantly lower than control plants.Studies have demonstrated that the VIGS system in Capsicum frutescens var had been successfully constructed,which provided a technology platform for large-scale validation of Capsicum frutescens var gene function. |
