摘    要:药物-血浆蛋白结合率是药动学研究的重要参数,通过超滤法结合高效液相色谱法测定了黄柏碱与血浆蛋白的结合率.含药血浆经超滤法得超滤液,以C18为固定相,乙腈-0.3%冰醋酸-三乙胺(体积比为20:80:0.5,其中0.3%为体积分数)为流动相,284nm检测波长下测定超滤液中黄柏碱.黄柏碱在0.20~20.00mg·L~(-1)质量浓度范围内线性关系良好,相关系数r为0.999 8,日内、日间精密度均小于2.5%.超滤法结果表明黄柏碱在不同浓度下的血浆蛋白结合率为26.45%~34.05%,蛋白结合常数为(1.214±0.132)×10~4 L/mol.

Abstract:Drug-plasma protein binding is an important parameter in pharmacokinetic studies. The protein binding of protein binding of phellodendrine with rat plasma was determined in this paper. A HPLC method was developed for the determination of phellodendrine in plasma ultrafiltrate. Chromatographic separation was achieved on a C18 column, using mobile phase consisting of acetonitrile-0.3% acetic acid- triethylamine (20:80:0.5, v/v/v) and UV detection at 284 nm. The standard curve for phellodendrine was liner over the concentration range of 0.2 to 20?g/mL with r=0.9998. The intra-day and inter-day variations were less than 2.5%. Ultrafiltration technique was applied to determining the plasma protein binding of phellodendrine in rat plasma. Results showed the plasma protein binding of phellodendrine was in the range of 26.45-34.05% over the all concentrations studied and the protein binding association constants was determined to be (1.214±0.132)×104 L/mol.
