单核细胞增多性李斯特菌对人、鼠脑微血管内皮细胞黏附、侵袭、增殖差异性分析

摘    要:为探究单核细胞增多性李斯特菌(LM)不同菌株对脑微血管内皮细胞黏附,侵袭以及在细胞内增殖的差异,本实验利用LM90SB2、LM3、LM4,塞氏李斯特菌(L.seeligeri),无害李斯特菌(L.innocua)感染脑微血管内皮细胞,采用BHI平板计数,测定细菌黏附率、侵袭率及不同时间细胞内增殖情况。结果表明:LM90SB2、LM3、LM4对脑微血管内皮细胞黏附率为3.14%~7.83%,统计学分析其差异不显著;LM90SB2、LM3、LM4对人/鼠脑微血管内皮细胞侵袭率(0.036%~0.128%)均大于L.seeligeri(0.0315%、0.00148%),L.innocua不侵袭脑微血管内皮细胞。LM90SB2、LM3、LM4对人/鼠2种细胞的侵袭率存在显著差异(P0.05),临床分离株LM90SB2增殖速度、菌量及持续时间明显高于其它菌株。以上结果提示LM不同菌株的毒力不同。

Abstract:To explore the Listeria monocytogenes strains on adhesion,invasion and the difference in cell profiferation in human/mouse brain microvascular endothelial cells,we used LM90SB2,LM3,LM4,L.seeligeri,and L.innocua to infect brain microvascular endothelial cells.The adhesion rate,invasion rate,and number of intracellular growth were measured by plate count method.The results showed that the adhesion rates of LM90SB2,LM3 and LM4 were ranged from 3.14% to 7.83%,with no significant statistical difference.The invasion rates of LM90SB2,LM3 and LM4 were varied from 0.036% to 0.128%,which were higher than L.seeligeri(0.0315%,0.00148%),and L.innocua was unable to invade BMEC.The invasion rates existed in significant difference(P〈0.05) in that three strains LM90SB2,LM3,and LM4 infected two cells,HBMEC and MNMEC,respectively.The clinical pathogenic strains LM90SB2 was higher than the others on the intracellular proliferation rate,maximum number and duration time in BMEC.These results indicated that different LM strains might be possessing different virulence.
