2-(2-(1H-1,2,4-三唑)乙基)-3-乙酰基-2,5-二取代芳基-1,3,4-噁二唑啉类化合物的合成

摘    要:在盐酸和乙醇存在下,苯乙酮、多聚甲醛以及N,N-二甲基甲酰胺进行Mannish反应,得到了Mannish碱;然后将其和1,2,4-三唑在水中反应,得到含有1,2,4-三唑的苯丙酮(1);在冰醋酸催化下,(1)和4-取代苯甲酰肼(2)进行缩合,得到一系列含有1,2,4-三氮唑的酰腙衍生物(3);化合物(3)在乙酸酐的作用下环化生成一系列2-(2-(1H-1,2,4-三唑)乙基)-3-乙酰基-2,5-二取代芳基-1,3,4-噁二唑啉(4).化合物(4)的结构经IR,1HNMR,MS,元素分析确证,同时用X-射线衍射法测定了化合物4a的晶体结构.

Abstract:In the presence of hydrochloric acid and ethanol, interaction of acetophenone, paraformaldehyde and N, N- dimethylformamide has Mannish reaction, and reactes with 1,2,4-triazole in water to get propiophenone with 1,2,4-triazole as its inclusion. Reaction of compound (1) with 4-substituted benzophenone hydrazide(2) gives corresponding hydrazones (3), which could be readily converted to their congener oxadiazolines (4) in good yields by treatment with acetic anhydride. Compounds(4) is confirmed by IR, ^1HNMR, MS, elemental analysis, Compounds 4a is determined by single-crystal X-ray diffraction.
