灵芝液体发酵产纤维素酶的提取及性质分析

摘    要:通过对比实验,找出了较适合灵芝液体发酵并产生纤维素酶的PDY培养基:马铃薯200g,葡萄糖20g,酵母膏5g,加水至1000mL,并对纤维素酶的酶活力进行了测定。发酵液经过硫酸铵盐析,纤维素酶被纯化了1.61倍,通过快速液相色谱的分离纯化,酶被纯化了1.96倍,酶活分别达到5.68和6.89。经过快速液相色谱分离纯化,有6个组分峰。其中第一、第二峰分离较好,含量较高。对灵芝纤维素酶的氨基酸组成进行了分析,灵芝纤维素酶并经紫外-可见光吸收光谱和红外吸收光谱对纤维素酶进行了分析。

Abstract:Through contrast experiment,the PDY medium which is fit for Ganoderma Lucidum's liquid fermentation and cellulase production is attained,including 200 g potatoes,20 g glucose,5 g yeast extract and 1 000 mL water.The enzyme activity of the cellulase from Ganoderma Lucidum is measured.The enzyme activities reachs 5.68 through salting out culture broth by(NH_4)_2SO_4 and 6.89 through fast protein liquid chromatography(FPLC),which is respectively 1.61-fold and 1.96-fold greater than 3.52 without using these methods in the process of isolation and purification of Ganoderma Lucidum cellulase.There are six components' peaks in FPLC spectrum.The spectrum shows that the isolation of the first peak and the second peak is better than that of the others and the components of these two peaks are higher.The amino acid components of Ganoderma Lucidum's cellulase are tested,and the characteristics of cellulase are analyzed by ultraviolet spectrum and infrared spectrum.
