一株红树尖瓣海莲内生真菌Penicillium sp.次级代谢产物及其生物活性研究

摘    要:采用活性追踪分离的方法,综合运用硅胶柱层析、Sephadex LH-20凝胶柱层析等,从红树尖瓣海莲(Bruguiera sexangula var.rhynchopetala)来源的一株青霉属真菌Penicillium sp.(3126)的发酵产物中分离鉴定了5个化合物.根据理化性质和NMR波谱方法等,并通过与文献对照,鉴定出化合物的结构分别为:麦角甾-22-烯-3β-醇(ergosta-22-triene-3β-ol)(1),麦角甾-5,7,22--烯-3β-醇(ergosta-5,7,22-triene-3β-ol) (2),过氧化麦角甾醇(ergosterol peroxide)(3),对羟基苯甲酸(4-hydroxybenzoic acid)(4)和对羟基苯甲醛(4-hydroxybenzaldehyde) (5).以上化合物均为首次从尖瓣海莲内生真菌中分离得到.活性测试表明,甾体类化合物1、苯衍生物4和5都具有较强的卤虫致死活性.

Abstract:Under the guidance of bioassay, column chromatography on silica gel and Sephadex LH-20 were used to isolate and purify the metabolites from the fermentation products of Penicillium sp. (3126), a fungus was obtained from a manBruguiera sexangula var. rhynchopetala grove. The structures of the isolated compounds were identified by spectroscopic analysis including NMR, and by comparison with the data of literature. Five compounds were isolated from the EtOAc extracts of 3126, and their structures were identified as ergosta-22-triene-3β-ol (I), ergosta-5,7,22-triene-3β-ol (2), ergosterol peroxide (3), 4-hydroxybenzoie acid (4) and 4-hydroxybenzaldehyde (5). All compounds were isolated from the fungus, which were isolated from the medicinal mangrove Bruguiera sexangula var. rhynchopetala for the first time. Compounds 1, 4 and 5 exhibited strong lethality to brine shrimp Artemia salina.
