五味子淫羊藿混合物提取工艺及其抗疲劳作用研究

摘    要:目的研究五味子与淫羊藿混合提取物(mixed Schisandra-Epimedium extract,MSEE)的最佳提取工艺,并观察其抗疲劳功效.方法采用加热回流提取法对五味子、淫羊藿进行混合提取,用正交实验法观察乙醇浓度(65%,70%,75%)、提取温度(60,65,75℃)、提取时间(1,1.5,2 h)及料液比(1∶10,1∶14,1∶18)对提取工艺的影响,并对提取物中五味子醇甲及淫羊藿苷含量进行分析,从中优选出最佳提取工艺.选取雄性ICR小鼠80只,随机分成空白对照组及150,300和600 mg/kg MSEE组,每组20只.给药30 d后,每组随机选取10只小鼠进行力竭游泳实验,记录小鼠力竭游泳时间.其余10只小鼠游泳90 min后处死,取血及肝组织,检测血清尿素氮及肝糖原水平.结果五味子与淫羊藿混合提取物最佳提取工艺:乙醇浓度为70%,提取温度为75℃,提取时间为1.5 h,料液比为1∶14.与空白对照组比较,300和600 mg/kg MSEE组小鼠力竭游泳时间均明显延长(P0.01);300和600 mg/kg MSEE组小鼠的血清尿素氮水平明显降低,而肝糖原水平则明显升高(P0.05或P0.01).结论得到MSEE最佳提取工艺,MSEE具有抗疲劳作用,可为开发具有抗疲劳功能的保健食品提供依据.

Abstract:Objective To study the optimal extraction process of the mixed extract of Schisandra chinensis and Epimedium sagittatum,and the antifatigue of the extract in vitro. Method The mixture of Schisandra chinensis and Epimedium sagittatum was extracted with heating reflux extraction. Orthogonal experiment effects of the ethanol concentration (65%,70% and 75%),the extraction temperature (60,65 and 75℃),the extraction time (1,1. 5 and 2 h) and the solid-liquid ratio (1 : 10,1 : 14 and 1 : 18) on the extraction process were observed, and the Schisandrin and Icariine contents were analyzed respectively,through which the extraction process of the mixed extract of Schisandra chinensis and Epimedium sagittatum was optimized. 80 male ICR mice were selected and randomly divided into control group,and 150,300 and 600 mg/kg MSEE groups,20 rats in each group. After the administration for 30 days,10 mice randomly selected from each group were for exhaustive swimming test,and the exhaustive swimming time of mice was recorded. The remaining 10 mice in each group were sacrificed after the swimming for 90 minutes,and their blood and liver tissue samples were taken for the detection of serum urea nitrogen and liver glycogen levels. Results The optimal extraction process of the mixed extract of Schisandrachinensis and Epimedium sagittatum was as follows: the ethanol concentration was 70%, the extraction temperature was 75℃,the extraction time was 1.5 h and the solid-liquid ratio was 1:14. Compared with those in the control group,swimming times of mice in 300 and 600 mg/ kg MSEE groups were significantly prolonged (P<0.01);serum urea nitrogen levels of mice in 300 and 600 mg/ kg MSEE groups were significantly lowered,and liver glycogen levels were significantly elevated (P<0.05 or P<0.01). Conclusion The optimal extraction process of MSEE was confirmed in this study. MSEE has an antifatigue activity. This study may provide the basis for the development of health food with antifatigue function.
