肿瘤细胞ECA109对甲醇等4种有机溶剂耐受剂量分析

摘    要:目的通过分析一定浓度下甲醇、乙醇、丙酮和甘油作用后肿瘤细胞ECA109细胞株形态和生长状态的变化,确定上述溶剂对该细胞株的相对安全剂量。方法按照10个浓度梯度(V/V)将甲醇等4种有机溶剂加入细胞培养基,作用后的12、24、36、48 h进行细胞形态观察和生存率检测。结果 4种有机溶剂作用于ECA109细胞后都表现出不同程度的细胞毒性作用,其中,乙醇的作用最为明显,20 mL/L作用12 h就出现明显的细胞毒性,细胞形态变化明显,生存率下降显著,10 mL/L时对细胞生长基本没有影响;甲醇和丙酮也有明显作用,30 mL/L以内甲醇的作用相对缓和,而20 mL/L以后丙酮的作用则增加明显,生存率下降的同时细胞形态也出现明显改变;甘油的作用最为缓和,并且仅抑制细胞生长而对细胞形态基本没有影响。结论 ECA109细胞对4种有机溶剂的耐受性有所不同,对乙醇的耐受剂量最低,其次为丙酮和甲醇,3种溶剂不仅抑制细胞的生长,还对细胞形态产生明显影响;甘油作用较为缓和,且仅对细胞株的生长产生抑制,并不导致细胞形态改变。

Abstract:Objective:To the morphologic changes and growth status of ECA109 were observed after being intervened by different concentrations of methanol/ethanol/acetone/glycerol to determine the safe dose of the four solvents. Methods: The morphologic changes of ECA109 cells were observed and the cell viabilities were also quantitated when cells were cultured for 12 h, 24 h, 36 h and 48 h respectively. Results: Four organic solvents all showed some cytotoxicity to ECA109 cell. Among them, ethanol showed the most obvious cytotoxicity. Except for growth inhibition, ECA109 also showed morphologic changes and low cell viability in 12 h at the concentration of 20 mL/L for ethanol, but ethanol at 10 mL/L almost had no effects on the cell growth. Methanol and acetone also showed obvious cytotoxicity to ECA109 cell and the effect of methanol was relatively less obvious within 30 mL/L, but acetone showed more effect on ECA109 cell growth and morphology if the concentration was above 20 mL/L. Glycerol also could repress ECA109 cell growth but had no effect on the morphology. Conclusion: ECA109 cells showed different tolerance to four kinds of organic solvents, the tolerance dose to ethanol was the lowest and next is acetone and methanol. They not only inhibited the cell growth but also caused the morphological changes. Glycerol only inhibited the cell growth but had no effect to cell morphology.
