细胞凋亡的三种形态学检测法的应用比较

文章比较分析了光学显微镜、荧光显微镜和扫描电子显微镜检测细胞凋亡的差异,探讨三种形态学检测法在尼古丁诱导的血管平滑肌细胞(vascular smooth muscle cells,VSMCs)凋亡中的应用。用终浓度为100μM的尼古丁作用于体外培养的大鼠血管平滑肌细胞24 h,不加尼古丁的细胞作为对照组,应用普通光学显微镜、荧光显微镜和扫描电子显微镜分别观察各组细胞的形态,确定尼古丁对血管平滑肌细胞凋亡的影响。三种形态学检测方法均可检测到尼古丁明显诱导血管平滑肌细胞的凋亡。荧光显微镜检测法因操作简便、灵敏度较高、价格适中,更适用于本实验。

Comparative study on three morphology testing methods for cell apoptosis

In the paper,the differences among three kinds of apparatus including light microscope,fluorescence microscope and scanning electron microscope in detecting cell apoptosis are compared.And the application of Vascular smooth muscle cells（VSMCs） apoptosis induced by nicotine is discussed.VSMCs isolated from rats are subjected to 100 μM nicotine for 24 hours,and the cells without nicotine are used to be compared.The morphology of apoptotic cells is observed by the light microscope,fluorescence microscope and scanning electron microscopy respectively.Typical cell apoptosis structure induced by nicotine is observed by the three methods apparatus.Because of the simple process,higher sensibility and lower price,the testing method by fluorescence microscope is more appropriate to the cell apoptosis.
