高效液相色谱-四级杆飞行时间串联质谱分析3种荷花的花色苷

摘    要:应用高效液相色谱-电喷雾/四极杆飞行时间串联质谱联用技术(HPLC-ESI/Q-TOF-MS/MS)分析了3种荷花(大红袍、洪湖红莲、红牡丹)的花色苷成分。首先以φHCl= 0.1%的甲醇超声提取荷花中的花色苷,然后用XAD-7大孔树脂纯化,选用Amethyst C18色谱柱(250 mm×4.6 mm,5 μm),以甲醇-水(φC2HF3O2=0.05%)为流动相进行梯度洗脱,柱后流出液采用电喷雾四极杆飞行时间质谱(ESI/Q TOF MS/MS)进行检测,共分离鉴定出6种花色苷,并且运用pH示差法分别测定了3种荷花中总花色苷的含量,为开发利用这一丰富的自然资源提供一定的参考依据。

Abstract:We employed HPLC-Electrospray Ionization/Quadrupole-Time of Flight-Mass/Mass Spectrometry(HPLC-ESI/Q-TOF-MS/MS) to analyze the anthocyanins in three species Nelumbo nucifera.The anthocyanins were extracted with methanol containing HCl(φHCl= 0.1%) and then purified on an Amberlite XAD-7 column.We applied an Amethyst C18 column(250 mm×4.6 mm,5 μm) to HPLC analysis with water methanol-water(containing trifluoroacetic acid(φC2HF3O2=0.05%)) in gradient as mobile phase.The eluates were then detected by ESI/Q-TOF-MS/MS.We altogether identified six species anthocyanins from these Nelumbo nucifera.We further determined their contents by pH-differential spectrophotometry.It provides theoretical foundation for further development and utilization of Nelumbo nucifera resources.
