多胺化壳聚糖载体固定化漆酶及其性质研究

摘    要:以多胺化壳聚糖为载体,戊二醛为交联剂固定化白毒鹅膏菌漆酶,研究了最佳固定化条件及固定化酶的部分性质.结果表明,戊二醛浓度2%,交联时间14h,固定化时间6h,给酶最70mg/g载体时同定化效果最佳,酶活力可达64.3 U/g.此固定化漆酶的Km为8.73mmol· L-1;最适pH值4.2,较自由漆酶向酸性偏移;最适反...

Abstract:Laccase has been immobilized on polyethyleneaminated chitosan with glutaraldehyde as crosslinking agent.When glutaraldehyde concentration is 2%,linking time is 14 h,immobilization time is 6 h and laccase dosage is 70 mg/g carrier the optimal condition was obtained.In this way 64 U/g of immobilized enzyme was available.The Km for the immobilized enzyme is 8.73 mmol·L-1 and the optimal pH and temperature for immobilized enzyme are 4.2 and 50 ℃.Compared with the free enzyme,its stability is considerable improved.
