Abstract:In this study,Aniline Blue Staining Method(ABSM) together with Tannic Acid Method(TAM) is utilized to screen the stain which is able to degrade lignin from the film of dead trees.The fungus is identified as Polyporus varius and its effects of pretreatment conditions on holocellulose content and kappa number in reed paper-pulping are studied.The results indicate that the optimum pretreatment conditions of Papermulberry by P.varius are shown as follows: pH 5.0,temperature 41℃,inoculum 50v/v,processing time 16d.At this point,the minimum kappa number is 17.851,with the reducing number of 2.236 compared with the control sample.Kappa number changes obviously,and the degradation of Phragmites australis holocellulose is not obvious,with the reducing degradation of 0.003%.Also,the degradation rate of lignin is investigated,and the degradation rate and delignification selectivity are 37.44% and 8.83,respectively.It indicates that the selected fungus has the ability to decompose lignin effectively and selectively.
