生物表面活性剂菌株的筛选及发酵条件初步优化

摘    要:从海南大学(儋州校区)职工食堂排污地采集的土样中分离到一株产生生物表面活性剂的菌株,命名为S423.采用16SrDNA系统发育学分析确定该菌株属于Klebsiella属.通过定性分析、薄层层析色谱(TLC)和红外分光光度法(IR)分析,活性物质初步鉴定为糖脂;通过正交实验初步优化该菌株的发酵条件;通过菌株S423发酵液优化前后的比较,发现优化培养基中发酵产生的表面活性剂活性高于基础培养基发酵产生的,排油圈直径增大3cm.

Abstract:A bacteria strain producing bio-surfactants was isolated from the sewage soil of staff eatery in Hainan University( Danzhou campus), and named as S423. The analysis results of 16S rDNA phylogenetic showed that the strain belongs to Klebsiella. The analysis results of TLC and IR indicated that the bio-surfactant of S423 was belonged to glycolipids. The orthogonal tests were used optimize the composition of the medium and fermentation conditions, and the compare results between different optimization conditions showed the activity of bio-surfac- tants from the optimized medium is higher than that from the non-optimized medium and the diameter of the oil circle is bigger 3cm.
