高寒草甸土壤放线菌分离方法研究

摘    要:采用平板涂抹分离法研究了培养基类型及样品处理对高寒草甸土壤中放线菌分离效果的影响。结果表明:改良HV琼脂和高氏1号加重铬酸钾(50mg/L)培养基是较好的分离培养基。土样在120℃于热处理1h后其悬浮液加0.05%SDS(十二烷基磺酸钠)和6%酵母膏,40℃振荡30min,能促进放线菌孢子萌发,增加放线菌的分离数量。

Abstract:The influence of medium type and different treatments of the soil samples fron alpine meadow on the isolation of aetinomyeetes was studied by plate paint isolation methods. The results showed that: the Gause' s starch agar added with 50 mg K2 Cr2O2/L medium and modified HV media were the best isolation medium of actinomyeetes. It might promote aetinomyeetes spore germination and increase the quantity of aetinomyeetes when the soils were treated with 120℃ for lh,after the soil suspension added with 6% yeast extract, 0.05% SDS and shaked at 40℃ for 30 min.
