摘    要:肿瘤转移是导致肿瘤患者死亡的最主要原因。细胞迁移在多步骤、多阶段的肿瘤转移过程中发挥着重要的作用。尽管有许多迁移相关分子的研究,但细胞迁移的分子机制仍不清楚。研究将xCELLigence细胞分析系统和RNA干扰技术结合,初步建立了实时无标记的肿瘤细胞迁移筛选技术体系。该技术体系的建立将为大规模的肿瘤细胞迁移筛选工作奠定基础,还将有助于发现肿瘤细胞迁移信号通路中新的调控分子,揭示肿瘤转移的分子机制。

Abstract:The majority of cancer deaths result from tumor metastasis which is a complex process involving several key steps. Cell migration is central to development and tissue remodelling and has a major role in cancer and metastasis. Increasing focus on genes regulate cell migration is apparent, whereas elucidation of the regulatory mechanisms requires additional studies. Here, the screening system for mammalian cell migration is established by combining the xCELLigence real-time cell analysis system and RNAi interference. Establishment of this system will make it feasible to perform a high throughput siRNA screening to identify genes involved in mammalian cell migration and help to reveal the mechanism of tumor metastasis.
