| 摘 要： | 从甲型肝炎病毒２个不同株感染的ＫＭＢ１７细胞中提取总ＲＮＡ，经ＲＴ－ＰＣＲ特异性扩增出此２毒株病毒蛋白２Ａ基因，将２个２Ａ基因克隆到ｐＧＥＭ－Ｔ载体上，经ＤＮＡ序列测定，得到２个２Ａ基因的核心苷酸序列，序列比较发现它们及与ＨＡＶ野毒株相对应序列有突变存在，Ｈ２株与ＨＭ１７５相比有一个点突变，同源性为９９．７２％；与ＨＭ１７５比较，Ｈ２株和Ｌ８株第３１９７位核苷酸均由Ａ突变为Ｇ，相应的氨基酸由丝氨酸

| Abstract: | The genes encoding viral protein enzyme 2A of two hepatitis A virus (HAV) strains (H 2 and L 8) were cloned with RT PCR with specific primers for 2A.The genes were sequenced after inserted into the vector pGEM-T.Sequence comparisons between the two strains and wild strain HM 175 showed that although there were mutations in 2A genes,similarity rates were high,99.37% between L 8 and HM 175 strain,99.72% between H 2 and HM 175 strain,99.65% between L 8 and H 2 strain.The effect of mutations in 2A on replication of HAV strains (H 2 and L 8) was also discussed. |
