甘农四号紫花苜蓿愈伤组织诱导研究

摘    要:以甘农四号紫花苜蓿的下胚轴、叶柄、子叶、叶片为外植体,研究了不同质量浓度的2,4-D对愈伤组织诱导的影响.结果表明:不同外植体的愈伤组织发生能力存在显著差异,外植体愈伤率下胚轴>叶柄>子叶>叶片.下胚轴在接种后16d的愈伤率高达100%,显著高于其他外植体.叶柄、子叶、叶片的愈伤率分别为:95.65%,88.06%,79.37%.其中叶柄的愈伤率显著高于子叶和叶片,子叶和叶片之间差异不显著.在附加3个不同质量浓度2,4-D的MS培养基上外植体的愈伤诱导率差异不显著,愈伤率均达到了90%以上.

Abstract:The research was conducted to explore the effect of dichlorophenoxyacetic acid(2,4-D)of different concentrations(1.5mg·L-1,2.0mg·L-1 and 2.5mg·L-1)for callus induction of hypocotyl,leaf blade,colyledon and stem of medicago sativa cv.Gannong No.4.The result showed that callus induction capacity varied significantly with the different explants,among which callus rate of hypocotyl was up to 100%,which was significantly higher than that of stem,colyledon and leaf blade,whose callus rates were 95.65%,88.06%,79.37%respectively.Callus rate of stem was significantly higher than that of colyledon and leaf blade,between which there was no significance observed.Among three media containing 1.5mg·L-1,2.0mg·L-1 and 2.5mg·L-1 2,4-D bear no differences in inducing callus formation of 4types of explants.
