H13诱导人卵巢癌A2780细胞凋亡及其机制的研究

摘    要:探讨新型Topo I抑制剂H13对人卵巢癌A2780的杀伤和诱导凋亡的作用机制.MTT法对人卵巢癌A2780的杀伤作用;流式细胞仪研究H13对A2780细胞的凋亡诱导作用;Western Blot法检测H13对A2780细胞内caspase-3和p53蛋白表达的影响.H13对人卵巢癌A2780有明显细胞毒性作用,并且有剂量依赖性;A2780细胞在H13作用后出现特征性凋亡形态特征,且凋亡率由5.7%上升为20.6%;H13明显增加p53蛋白的表达和caspase-3蛋白的表达.H13对人卵巢癌A2780有明显的杀伤和促凋亡作用,其机制可能依赖于细胞内p53蛋白和caspase-3蛋白表达增高.

Abstract:To investigate the cytotoxicity and proapoptotic activity of H_(13) on cisplatin-resistant human ovarian cancer cell lines A_(2780) and its mocular mechanisms.The cytotoxicity of H_(13) on A_(2780) was monitered using MTT assay; Flow cytometry was employed to observe the apoptosis of A_(2780) induced by H_(13);Western blot were used to determine the expression of p53 and caspase-3.H_(13) had strong cytotoxicity on A_(2780) cell invitro,and the cytotoxicity was dose-dependent. H_(13)-treated cells demonstrated apoptosis property while the apoptotic rate was increased from 5.7% to 20.16%.The expression of p53 were upregulated and the expression of caspase-3 in the course of A_(2780) apoptosis when treated with H_(13).The cytotoxicity and proapoptosis activity of H_(13),which is significantly observed in cisplatin-resistant human ovarian cancer,may be related to the increased p53 expression and caspase-3 expression.
