摘    要:采用染料亲和层析、QAE离子交换层析等纯化步骤,从绝经期妇女尿促性腺激素(HMG)粗品中分离纯化获得高纯度卵泡刺激素(FSH)产品,其中FSH的生物活性达到268.1IU/mg,卵泡刺激素与黄体生长素(LH)的活性比值达到259,产品超过欧洲药典对高纯度卵泡刺激素的要求。此工艺FSH的活性回收率高达65%,而且分离纯化步骤少,简单易行,适合于大规模生产。

Abstract:High purity follicle stimulating hormone(FSH)was produced from human menopausal gonadotropin(HMG)crude extracts by utilizing a new purification design in which dye affinity chromatography(DAC)along with QAE ion_exchange chromatography was used to greatly enhance the purity and bioactivity. The bioactivity of the FSH product reached 268.1IU/mg and the ratio between FSH and leuteinizng hormone(LH)amounted to 259, which allowed for the FSH product to comply with the UP standards set for high purity FSH products. With a total recovery rate as high as 65% for the FSH bioactivity,this purification process will be applicable to large scale manufacturing of FSH.
