摘    要:本文对天然鸡血SOD中的金属辅基进行了去除和重组,初步研究了Cu~(2+)、Zn~(2+)、Co~(2+)等二价阳离子对SOD活性的影响。对天然态Cu_2Zn_2-SOD、重组Cu_2Co_2-SOD及脱辅E_2E_2-SOD的活性测定和光谱学行为的分析表明,重组Cu·Co-SOD具有同天然Cu·Zn-SOD类似的酶活性。在pH3.8时加入Cu~(2+)和Co(2+),重组的金属酶活性可恢复至天然酶活的85%,紫外吸收图谱也基本一致。

Abstract:The removal and reconstitution of metal prosthetic groups in native chicken crythrocyte superoxide dismutase is reported. The effects in enzyme activity and function of metal-substituted derivatives (Cu~(2+), Zn~(2+), Co~(2+)) are investigated. It is indicated that the reconstituted Cu_2 Zn_2-SOD has the same enzyme activity as that of native Cu_2 Zn_2-SOD, by the determination of activity and analysis of spectral behaviours on Cu_2Zn_2-SOD, Cu_2 Co_2-SOD and E_2E_2-SOD. When adding Cu~(2+) and Co~(2+) to Apo-SOD at pH 3.8, its catalytic activity could be restored to 85% and the UV Spectra also recovered.
