| 摘 要： | 利用磷酸钙沉淀法将重组的质粒ｐＣＭＶ５－ｐｅｍｔ２与ｐＳＶ－ｎｅｏ共转染，经Ｇ４１８筛选免疫细胞化学和免疫印迹法检测蛋白表达。获得稳定表达ＰＥＭＴ２的阳性克隆。

| Abstract: | The inhibitory effect of phosphatidylethanolami ne N-methyltransferase 2(PEMT2) on the growth of CBRH-7919 hepatoma cells(AFP po sitive) is studied.ABC methods and acti vity assay show that PEMT2 gene is successfully expessed in hepatoma cells CB RH 7919.Cell cycle analysis shows that the proportions of hepatoma cells pCMV 5-pemt2 CBRH-7919 in G1 phase increases and in S phase decreass in comparison wi th those of parallel cell lines. MTT assay shows that proliferation of hepato ma cells is reduced markedly and content of AFP is lowered too.Expessi on of PEMT2 may inhibit the growth of hepatoma cells,which shows that PEMT2 gene is a suppressor gene. |
