不同S180细胞株蛋白质特性的电泳及免疫组化法比较研究

用ＳＤＳ聚丙烯酰胺凝胶电泳（ＳＤＳ－ＰＡＧＥ）和免疫组化染色方法，比较研究４个不同单位保种的小鼠肉瘤（Ｓ１８０）细胞蛋白质的表达。电泳结果显示，北京市肿瘤研究所仲种的Ｓ１８０蛋白质条带数最多，武汉大学保种中心的Ｓ１８０最小。免疫组化染色法测定四个单位Ｓ１８０细胞的Ｒａｆ－１、Ｔｒｋ、Ｇａｉ、Ｇａｏ和ＮＦＫｂＰ６５蛋白表达，Ｇａｉ、ＴｒｋＡ和Ｒａｆ－１阳性率均低于０．２％；Ｇａｏ阳性率均低于４．２％

Comparision of Protein Distribution in Different S180 Strains with Electrophoresis and Immunohistochemistry

To study the protein distribution in different S180 Strains using SDS polyacrylamide gel electrophoresis (SDS-PAGE) and immunohistochemistry were used.The number of protein bands of Sarcoma 180 strain from The Beijing Cancer Institute was the most and Wuhan University's was the least.The protein distribution of Raf\|1,TrkA,Gai,Gao and NF k Bp65 were observed in different S180 strains with immunohistochemistry.The positive rate of TrkA,Gai and Raf\|1 was less than 0.2%.Positive rate of Gao was 4.2%. The positive rates of NF k Bp65 were 76.0%,23.1%,4.5% and 17% respectively.Chitest showed significantly difference (P<0.01) excepting between S180 strains from Institute of Pharmacology of Chinese Medical Academy and those from Beijing Cancer Institute.The results indicate that some protein compositions of the Sarcoma 180 cells from 4 institutes not only have quantitative,but also have qualitative differences.Therefore the quality of S180 cell strain should be controlled.
