杜氏藻与大肠杆菌融合初探

摘    要:杜氏藻(Dunaliella peircei Nicolai)系真核单胞绿藻,无纤维素细胞壁,细胞膜外由糖蛋白类物质组成柔软的包被,E.coli(Cm~r)内含带Cm~r基因的质粒。以对氯霉素(Cm)抗性的变化作为杜氏藻和大肠杆菌融合的选择标记,因而首先测定了它们在各自琼脂培养基上对Cm(原始浓度125g/1)的抗性水平(Tab.1)。

Abstract:The protoplasts of D, peircei (Chlorophyceae, Vohocales) can be obtained by inducing with pronase digestion. They were fused with the spheroplasts of E. Coli(Cmr) by polyethylene glycol(PEG) method. Some Dunaliella-like colonies were isolated from the selection media,with the chloramphenicol(Cm) resistibility rising from 70 ppm to 150 ppm. E. coli(Cmx) could still be found in the cells of fusants two weeks after fusion.
