摘    要:目的通过模拟现实环境,对国家药物安全评价监测中心可能使用的4种处理动物饮水的储存时间-菌落进行监测,为该中心的实验动物饮水管理提供参考。方法将4种水样存放在模拟现实环境下,每周分别取样接种血平皿进行细菌培养,为控制样品传递可能造成的污染,取样及接种操作安排在临近储存环境的设备中进行。结果高压灭菌管道自来水连续5周未检测到细菌,高压灭菌过滤水连续8周未检测到细菌;新鲜未高压灭菌管道自来水及过滤水水样细菌检出阳性,其中新鲜自来水存放1周后每毫升水样中菌落单位超过100个,新鲜过滤水存放1周后每毫升水样中菌落单位平均34个(范围:29~37 cfu/mL)。结论高压灭菌水储存5周满足无菌要求,可提供给清洁级以上动物饮用;未经高压灭菌过滤水储存1周内可满足国标要求,可供普通级动物使用,但接取新鲜水样将更加安全;新鲜管道自来水满足国标要求,可供普通级动物使用,但建议不要储存过夜饮用。

Abstract:Objective A series of bacterial culturing was conducted for water samples collected at different time points during storage, in order to develop and fine-tune, based on science, the animal drinking water management policy at National Centre for Safety Evaluation of Drugs (NCSED). Method Animal drinking water samples treated with 4 different methodologies were stored in mimicked conditions which reflecting the real practices, Water sampled weekly after water storage was inoculated onto blood agar plates for bacterial culturing. The sampling and inoculating were conducted aseptically in equipments near the storage rooms to minimize the risk of contamination.Result The culturing of the autoclaved piping tap water, and autoclaved filtered water samples, collected in the first 5 and 8 weeks, respectively, post storage did not reveal any colony growth. Colony growth was observed in the fresh piping tap water and fresh filtered water samples. The cultured piping tap water sampled after 1 week of storage had more than 100 cfu/mL (colony-forming units per milliliter) , whereas the filtered water sampled also at 1 week post storage had an average of 34 cfu/mL (range:29 -37 cfu/mL). Conclusion Under the experimental conditions (mimicking the real practice), the autoclaved water stored up to five weeks can meet the sterile requirements, and be provided to clean and above level of animals for drinking. Although the fresh filtered water is safer, the non-autoclaved filtered water stored within 1 week can meet the requirements of GB 5749 -2006 and GB 50447- 2008, and it is ok for conventional animal consumption. Fresh piped tap water can meet the GBs requirements for conventional animal drinking water, however it is not recommended to store over night.
