大豆蛋白酶解物抗氧化及促进微生物生长研究

摘    要:研究Alcalase蛋白酶对大豆蛋白的水解作用,通过正交设计确定酶解最佳条件,探讨最佳水解条件下不同酶解时间酶解物的抗氧化性和促进微生物生长的作用.结果表明,酶解最佳条件为:pH 9.0、温度65℃、底物酶量比100 g/mL;酶解90 min的酶解物清除自由基能力最强,酶解30 min的酶解物对酵母菌生长有促进作用,酶解60 min酶解物对酵母菌的代谢作用显著;酶解60、90 min的酶解物对黑曲霉的生长均有较好的促进作用.可见,酶解时间不同,酶解物的抗氧化活性与促进微生物生长作用有差别.

Abstract:Hydrolysis effect of soybean protein with Alcalase proteinases was studied,and the optimal hydrolysis conditions were selected by orthogonal design.Under the optimal hydrolysis conditions,the antioxidant activity and the acceleration effect to the growth of microorganism in different hydrolysis time of soybean isolated protein enzymatic hydrolysates(SEH) had been discussed.Results showed that the hydrolysis condition pH of 9.0,temperature of 65 ℃,the ratio substrate and enzyme dose of 100 g/mL was the optimum;The SEH with hydrolysis time of 90 min eliminated the hydroxyl radicals best;The SEH with hydrolysis time of 30 min could accelerate the growth of Saccharomyces carlsbergensis,and the SEH with hydrolysis time of 60 min could accelerate the metabolism effect of Saccharomyces carlsbergensis obviously;The SEH with hydrolysis time of 60 and 90 min could accelerate the growth of spergillus flavus preferably.It is shown that the SHE under different hydrolysis time has different antioxidant activity and acceleration effect to the growth of microorganism.
