摘    要:以细胞浓度为菌体生长指标,探索并优化对氯甲苯降解菌株Bacillus licheniformis ycsd01 种子培养基中碳源、氮源、主要无机盐等培养基组分及其用量,最终获得菌株适宜的培养基配方.结果表明:对氯甲苯降解菌株的最佳种子培养基配方为2.0g·L^-1对氯甲苯,8.0g ·L^-1葡萄糖,2.5 g·L^-1 NH4Cl,4.5 g·L^-1 K2HPO4,2.0 g·L^-1 KH2PO4,0.2 g·L^-1 MgSO4,0.05 g·L^-1MnSO4,0.01 g·L^-1 FeSO4· 7H2O和0.03 g·L^-1 CaCl2·2H2O.菌株在优化后较优化前的生长曲线延滞期缩短,指数期和稳定期变长,更晚地步入衰亡期,各生长时期的细胞数量均显著增加,有望用于大规模生产.

Abstract:Taking the cell concentration as the cell growth indicator,the suitable formulations of the seed enlarge medium for the p- chlorotoluene degrading strain Bacillus licheniformis ycsd01 was obtained by exploring and optimizing the medium components and amounts such as carbon and nitrogen sources and main inorganic salts. The results show that the suitable formulations of the seed enlarge medium for the p- chlorotoluene degrading strain were 2. 0 g·L- 1p- chlorotoluene,8. 0 g·L- 1glucose, 2. 5 g·L- 1NH 4 Cl,4. 5 g·L- 1K 2 HPO 4,2. 0 g·L- 1KH 2 PO 4,0. 2 g·L- 1MgSO 4,0. 05 g·L- 1MnSO 4,0. 01 g·L- 1FeSO 4 ·7H 2 O and 0. 03 g·L- 1CaCl 2 ·2H 2 O. Optimization comparing to no optimization shortened the lag phase but prolonged the exponential phase and stationary phase of the growth curve and entered later into the decline phase. The cells concentration in each growth period was significantly increased and expected for mass production.
