洁霉素产生菌原生质体的形成、再生和融合

摘    要:洁霉素产生菌不论生长在含有甘氨酸的S培养基或不含甘氨酸的S培养基的菌丝中,其细胞壁均可被溶菌酶溶解而释放出原生质体。甘氨酸浓度为1.0%破壁效果最好,溶菌酶的作用浓度以2.5mg/ml为最佳。原生质体在Hb培养基上能够很好地再生。用P培养基配制的40%(w/v)PEG1000溶液,能有效地诱导洁霉素产生菌原生质体融合。融合重组频率达6.7%。

Abstract:This paper studies the formation, regeneration and recombination of proto- plasts from the mycelia of lincomycinum productive strain which grown on S medium containing glycine or not. The cell wall of mycelia can be solved by ly- sozyme and thus protoplasts liberated. 1.0% glycine in S medium and 2.5mg/ml lysozyme are the optimum concentration for its liberation. The protoplasts rege- nerate very well on Hb medium. The protoplasts recombinate effectively by fu- sion in 40% (w/v) PEG 1,000 solution prepared with P medium, with a recom- bination ratio of 6.7%.
