MiR-199b-3p、miR-373、miR-548c-3p及Lin28在肺癌中表达及其意义

摘    要:探讨miR-199b-3p、miR-373、miR-548c-3p与Lin28在肺癌中的表达,以及它们与临床指标的相关关系.用溶液法提取46例肺癌患者的癌组织和癌旁组织中的总RNA,Q-PCR检测基因的相对表达量.经过两独立样本t检验,比较miR-373(t=3.28,P=0.000 7)、lin28(t=3.94,P=0.001 6)及miR-548c-3p(t=2.98,P=0.002)在肺癌组织和癌旁组织中的表达,基因表达均有显著性差异.使用Spearman肺癌组织中RNA与免疫组织化学指标发现MiR-b199b-3p与MRP(Rs=0.40,P=0.029),miR-548c-3p和Ki167呈正相关关系(Rs=0.52,P=0.0056),miR-373与K167呈正相关关系(Rs=0.43,P=0.029),Lin28与P53呈负相关关系(Rs=-0.43,P=0.022),还与PGP呈负相关关系(Rs=-0.38,P=0.034).miR-373在肺癌患者组织中基因表达显著下调,而miR-548c-3p与Lin28基因表达呈上调,3个基因的表达与肿瘤的发生发展关系密切.

The expression characteristics of miR-199b,miR-373, miR-548c and Lin28 in lung neoplasm

Abstract:This research investigates the expression of miR-199b-3p, miR-373, miR-548c-3p and Lin28 in lung neoplasm and their correlation with clinicopathological parameters. The total RNA was extracted from TRIzol in 46 cases of lung neoplasm and its adjacent tissues. The relative expression of genes was detected by real-time fluorescence quantitative PCR. After two t-tests of independent samples, it compared the expression of miR-373(t=3.28, P=0.000 7), Lin28(t=3.94, P=0.001 6) and miR-548c-3p(t=2.98, P=0.002) in lung neoplasm and the adjacent normal tissue, and found that the expression of gene was significantly different. There was a positive correlation between miR-b199b-3p and MRP(Rs=0.40, P=0.029), alcohol consumption(Rs=0.32, P=0.043), and miR-548c-3p and Ki167 in Spearman's lung cancer tissues and immunohistochemistry(Rs=0.43, P=0.029). There was a negative correlation between Lin28 and P53(Rs=-0.43, P=0.022), but also a positive correlation between the miR-373 and K167, and a negative correlation with PGP(Rs=-0.38, P=0.034). MiR-373 is down-regulated in lung neoplasm, and miR-373 may inhibit tumorigenesis and progression of lung neoplasm through the tumor suppressor gene during the development of lung neoplasm. While the expression of miR-548c-3p and Lin28 was up-regulated, miR-548c-3p and Lin28 may play an important role in the progression of lung neoplasm through inhibiting the expression of suppressor genes in lung neoplasm.
