经典连苯三酚SOD测活法(420nm法)的某些影响因素

摘    要:本文对超氧化物歧化酶(SOD)的经典连苯三酚测活法的某些影响因素进行了考查。结果发现:固定自氧化速率,pH值改变0.10单位将引起20%左右的测定误差;固定连苯三酚终浓度,同样程度的pH变化仅产生约13%的测定误差。在低浓度下(最终浓度小于0.15mmo1/L),连苯三酚的微小变化会严重影响活性测定结果。

Abstract:This paper examined some factors influencing superoxide dismutase(SOD) activity assay in the classicai pyrogallol method(420nm method).The resultsshowed that when the autoxidation rate was kept constant in the assay system,the pHchange of 0.10 unit could result in 20% error of the enzyme activity measured.If the finalconccntration of pyrogallol was kept constant,the pH change of same unit resulted in13%error of the activity. At low concentration of pyrogallol,say less than0.15mmol / L,the very little change of pyrogallol concentrations could seriously affectthe results of the assay.
