摘    要:目的 :研究电针对脑梗死大鼠尾壳核、海马CA1区缺血半影 (IP)区神经元型一氧化氮合酶 (nNOS)免疫阳性神经元的影响 ,为探讨针刺治疗脑梗死神经机制提供实验依据。方法 :采用线栓法建立大鼠永久性大脑中动脉栓塞的脑梗死模型 ,分为缺血组和缺血 +电针组 ;用免疫组化的方法观察尾壳核、海马CA1区IP区nNOS免疫阳性神经元表达的变化。结果 :(1)缺血组 :缺血 1d ,尾壳核IP区nNOS免疫阳性神经元深染固缩 ;缺血 7d后 ,阳性神经元数目增加 (P <0 0 5 ) ;在海马CA1区IP区 ,缺血 1d ,阳性神经元分布广 ;缺血 7d ,数目减少 (P <0 0 5 ) ;缺血 14d ,数目较缺血7d时增加 (P <0 0 5 )。 (2 )缺血 +电针组 :尾壳核IP区nNOS免疫阳性神经元数目较缺血组增加(P <0 0 5 ) ;在海马CA1区IP区 ,电针 1d ,阳性神经元分布局限 ,数目较缺血组减少 (P <0 0 1) ,电针 7d后 ,数目较缺血组增多 (P <0 0 5 )。结论 :电针对nNOS免疫阳性神经元调节具有双重性。既能减少电针 1d时海马IP区nNOS表达 ,减轻一氧化氮 (NO)的毒性作用 ;又能加速海马 (电针 7、14d)、尾壳核IP区nNOS表达的恢复

Effect of electroacupuncture on the expression of NOS-positive neurons of cerebral infarction rat
Abstract:To observe the effect of the electroacupuncture on the expression of nNOS-positive neurons of cerebral infarction rat in the ischemic penumbra, and to supply experimental evidence for exploring central nervous mechanism of the electroacupuncture in ischemic penumbra of caudate putamen, hippocampal subfield CA1. Methods: Establishing rat model of cerebral infarction with the middle cerebral artery occlusion, the rats were divided into two groups: ischemic group and the electroacupuncture-treated group, the change of expression of NOS-positive neurons was observed in caudate putamen, hippocampal subfield CA1 with immunohistochemistry. Results: (1) Ischemic group: The staining of the bodies of nNOS-positive neurons was deep, the size decreased at 1st d with ischemia, the number increased after 7 d with ischemia in caudate putamen (P<0.05). The staining of the bodies of nNOS-positive neurons was deep, the distribution was widespread at 1st d with ischemia, the number decreased obviously at 7th d with ischemia (P<0.05), the number was more at 14 th d than at 7 th d with ischemia in hippocampal subfeild CA1 (P<0.05). (2) The electroacupuncture-treated group: the number was more than ischemic group caudate putamen (P<0.05). The number was less than ischemic group at 1st d with electroacupuncture (P<0.01), and more than ischemic group after 7 d with electroacupuncture in hippocampus (P<0.05). Conclusion:The regulation of the electroacupuncture to NOS-positive neurons had two patterns: It decreased the expression of NOS-positive neurons at 1st d of ischemia period in hippocampal subfield CA1, decreased the toxicity of NO; it also promoted the restoration of nNOS in caudate putamen and hippocampal subfield CA1(at 7,14 d with ischemia).
