榄香烯乳对B16细胞株的诱导分化作用

摘    要:观察榄香烯乳对B16黑色素瘤细胞株是否具有诱导分化作用。通过MTT显色法测定榄香烯乳在体外对B16细胞株的IC50,腹腔给药观察榄香烯乳对体内B16细胞株皮下、肺转移结节的抑制作用.选取低于IC50的药物浓度研究榄香烯乳作用后B16细胞株超微结构和成瘤性的改变.榄香烯乳对体内外B16细胞株均有抑制作用.药物作用后,瘤细胞生长抑制,增殖减慢,接触抑制恢复,成瘤性降低,电镜观察细胞恶性度下降,出现较为成熟的黑色素颗粒.在低浓度时,榄香烯乳对B16细胞株具有诱导分化作用

Abstract:To determine if elemene emulsion induces B16 cells differentiation. The MTT method was used for the determination of IC50 of B16 cells by elemene emulsion. The inhibition of the subcutaneous and lung metastasis tumor in mice was observed by injected intraperitoneally elemene emulsion. we studied the ultrastruture and tumorigenesis of B16 cells in low drug dosage. Both in vitro and vivo B16 cells were inhibited by elemene emulsion. Besides, the ultrastructure showed the morphological changes, such as microvilli decrement, nucleus pyknosis, typical melanogen. Signs of induction of differentiation were observed in low dosage.
