南川百合组织培养与快繁技术体系研究

摘    要:为加快南川百合的繁殖速度,对南川百合进行离体组织培养研究,结果表明:南川百合外植体的诱导率从大到小依次为:鳞片,花丝,子房,茎段,叶片.用鳞片作为外植体时,下部的诱导率最高,中部次之,上部最差.鳞片诱导丛生苗的最佳培养基为MS+6-BA 1.5mg/L+NAA 0.5mg/L;用花丝和子房作为外植体时,诱导丛生苗的最佳培养基为MS+6-BA 0.5mg/L+NAA 0.5mg/L;用再生苗的叶片作为外植体时,诱导丛生苗的最佳培养基为MS+6-BA 1.0mg/L+NAA 0.5mg/L;用再生苗的茎段作为外植体时,诱导丛生苗的最佳培养基为MS+6-BA 1.5mg/L+NAA 1.0mg/L.用于不定芽增殖的最佳培养基为MS+6-BA 1.0mg/L+NAA0.05mg/L.无根苗生根的最佳培养基为1/2MS+NAA 0.5mg/L,生根率可达到86.7%,移栽后成活率很高.

Abstract:This research employed tissue culture to accelerate the speed of propagation of Lilium rosthornii Diels and shorten its reproductive cycle.The induction rate of L.rosthornii explants appeared to be in the order of scale>filigree>ovary>stem segment>leaf.With the scales as the explants,their lower part had the highest induction rate,followed in sequence by the middle part and the upper part.The best induction medium for clustered shoots from the scales was MS+6-BA 1.5 mg/L+NAA 0.5 mg/L.With filigree and ovary as the explants,the best medium was MS+6-BA0.5 mg/L+NAA0.5 mg/L.With the leaves and the stem segments of the regeneration plants as explants,the best medium was MS+6-BA1.0 mg/L+NAA0.5 mg/L and MS+6-BA1.5 mg/L+NAA1.0 mg/L,respectively.The best medium for adventitious bud proliferation was MS+6-BA1.0 mg/L+NAA 0.05 mg/L.The best rooting medium was 1/2MS+NAA 0.5 mg/L,which gave a rooting rate as high as 86.7%,and the survival rate was high after transplanting.
