| 摘 要： | 以水提醇沉法从牛膝中提取得到牛膝多糖,采用苯酚-硫酸法测定其多糖含量,以正交实验优化显色条件.确定5%苯酚用量1.0mL,浓硫酸用量7.5 mL,反应时间30 min.标准曲线回归方程为A=0.057 2C-0.006 9,r=0.9991(n=7),在2.5～30.0mg/L范围内呈良好线性关系.平均回收率为97.5%,RSD=1.69%(n=5).测得多糖平均含量为29.4%,RSD=1.84%(n=6).该法简单、准确、稳定,可为其继续研究开发及应用提供参考依据.

| Abstract: | The roots of Achyranthes bidentata.was defatted with 80% EtOH,extracted with water and precipitated with 80% EtOH,and then was deproteined by Sevag method,and from which,a crude polysaccharide noted as ABPS was obtained.The content of ABPS was determinated by phenol-sulfuric acid method.The main fators affecting the method were optimized and the results showed that the optimal conditions were the time of reaction of 30min and the dosage of 5% phenol and H2SO4 of 1.0 and 7.5 mL,respectively.Linear range was ... |
