摘    要:在5L小型发酵罐上,考察了不同搅拌转速对细胞生长和碱性果胶裂解酶生产的影响。根据细胞生物量、酶活水平、比细胞生长速率和比产物合成速率的变化情况,提出了两阶段搅拌转速控制策略:0~5h搅拌转速500r/min;5h后搅拌转速600r/min。PGL和PIVIGL酶活分别提高5.3%和15.1%;FGL和PMGL最大平均比生成速率分别提高15.9%和23.1%。

Abstract:Batch enzyme fermentation by Bacillus subtilis at different agitation rate ranged from 400 r/min to 600 r/min was studied in a 5 L fermentor. Based on time courses of the specific cell growth rate and specific alkaline pectate lyase production rate, a two-stage agitation rate control strategy was developed, in which agitation rate was controlled at 500 r/min at the first 5 h, then switched to 600 r/min afterwards. Applying this two-stage agitation rate control strategy, E_(1max) and E_(2max) could be improved by 5.3% and 15.1%, (?)_(1max) and (?)_(2max) could be improved by 15.9% and 23.1%, respectively.
