酵母诱导子对肉苁蓉细胞悬浮培养影响的研究

摘    要:目的:以酵母诱导子对肉苁蓉细胞悬浮培养及其抗氧化活性成分苯乙醇苷的合成动力学的影响进行研究.方法:从肉苁蓉种子诱导出愈伤组织,在MS培养基上进行细胞悬浮培养.首先考察肉苁蓉细胞生长和苯乙醇苷合成的动力学,在此基础上就酵母诱导子对细胞生长和苯乙醇苷合成的影响进行研究.结果:肉苁蓉悬浮细胞培养经过三天的延滞期后进入对数生长期,21天后细胞生长进入稳定期,最大生物量和苯乙醇苷产量分别达到8.03g DW/L和97 mg/L.在对数生长期后期,即培养的第18天加入0.8 mL/瓶浓度的酵母诱导子,诱导三天的效果最好,苯乙醇苷的最高产量为237.7 mg/L,为对照的239%.结论:合适浓度的酵母诱导子不但能够有效提高肉苁蓉细胞悬浮培养液中的苯乙醇苷含量,同时对细胞生长也具有促进作用.

Abstract:The purpose of the study is to investigate the kinetics of cell growth and PeG accumulation in the medium and effect of yeast elicitors on cell growth and PeG synthesis. The results show that in C.deserticola suspension cell cultures, cells began to grow exponentially after three-day lag phase, and reached stationary phase on day 21. The maximum biomass and PeG production of C.deserticola cell are about 8.03g DW/L and 97mg/L respectively. Not only PeG content but also biomass level have been enhanced by addition yeast elicitor in C.deserticola suspension cell cultures. When 0.8mL/flask of yeast elicitor is added to the medium on day 18 with the three days of inoculum, the maximum PeG production(237.7mg/L) is obtained, an increase of 239%.
