| 摘 要： | 比较序列特异性引物聚合酶链反应(PCR-SSP)和流式磁珠反向序列特异性寡核苷酸杂交法(reverse sequence specific oligonucleotide,RSSO)在造血干细胞捐献者资料库四川分库捐献者DNA的HLA基因分型中的应用优劣.对造血干细胞捐献者资料库109份样本同时用RSSO和PCR-SSP方法检测,并分析HLA-ABDR结果.109份样本用流式磁珠反向SSO杂交法所检测的HLA-ABDR分型一次成功率为100%;而PCR-SSP方法所检测的HLA-ABDR分型结果合格率为约98.1％.RSSO和PCR-SSP两种方法均可用于骨髓库样本检测,但侧重不同.

| Abstract: | To investigate the advantages and disadvantages of PCR-SSP and SSO method in Sichuan bone marrow donors HLA typing. A total of 109 samples were typed by SSO and SSP.All of typing results were analyzed and compared.109 samples were correctly typed by PCR-SSO.While the ratio of wrong HLA typing by SSP method was 1.9%.Both RSSO and PCR-SSP methods can be applied in HLA typing of bone marrow donors. |
